Automated Techniques for Stability Analysis of Delta-Notch Lateral Inhibition Mechanism
نویسندگان
چکیده
We describe, in detail, the modeling and automatic analysis of the Delta-Notch lateral inhibition mechanism using a hybrid systems framework. Inhibitory lateral signaling between adjacent cells is one of the central processes responsible for cell-differentiation in a cluster of identical cells. The basic modeling unit used in this paper is a hybrid automata and more complex models are built using compositions over hybrid automata. Analysis of the resulting model is done by creating a finite-state discrete abstract transition system for the hybrid model and model-checking this abstraction against the properties of interest. The abstraction technique, which is completely automated, uses suitable support from decision procedures and theorem-provers. The automated abstraction of continuous models of biological systems onto discrete transition systems enables new kinds of analysis on problems of biological interest.

Mutual inactivation of Notch and Delta permits a simple mechanism for lateral inhibition patterning
Lateral inhibition patterns mediated by the Notch-Delta signaling system occur in diverse developmental contexts. These systems are based on an intercellular feedback loop in which Notch activation leads to down-regulation of Delta. However, even in relatively well-characterized systems, the pathway leading from Notch activation to Delta repression often remains elusive. Recent work has shown t...
متن کاملPattern formation by lateral inhibition with feedback: a mathematical model of delta-notch intercellular signalling.
In many developing tissues, adjacent cells diverge in character so as to create a fine-grained pattern of cells in contrasting states of differentiation. It has been proposed that such patterns can be generated through lateral inhibition--a type of cell-cell interaction whereby a cell that adopts a particular fate inhibits its immediate neighbors from doing likewise. Lateral inhibition is well ...
متن کاملRobust selection of sensory organ precursors by the Notch-Delta pathway.
The patterning of multicellular organisms is robust to environmental, genetic, or stochastic fluctuations. Mathematical modeling is instrumental in identifying mechanisms supporting this robustness. The principle of lateral inhibition, whereby a differentiating cell inhibits its neighbors from adopting the same fate, is frequently used for selecting a single cell out of a cluster of equipotent ...
متن کاملAn Expanded Notch-Delta Model Exhibiting Long-Range Patterning and Incorporating MicroRNA Regulation
Notch-Delta signaling is a fundamental cell-cell communication mechanism that governs the differentiation of many cell types. Most existing mathematical models of Notch-Delta signaling are based on a feedback loop between Notch and Delta leading to lateral inhibition of neighboring cells. These models result in a checkerboard spatial pattern whereby adjacent cells express opposing levels of Not...
متن کاملMultiscale analysis of pattern formation via intercellular signalling
Lateral inhibition, a juxtacrine signalling mechanism by which a cell adopting a particular fate inhibits neighbouring cells from doing likewise, has been shown to be a robust mechanism for the formation of fine-grained spatial patterns (in which adjacent cells in developing tissues diverge to achieve contrasting states of differentiation), provided that there is sufficiently strong feedback. T...
